Determination of macroelements, organic acids and other parameters in ketchups

The aim of this master’s thesis is perform basic analyses of samples ketchups. The theoretical part describes the basic characteristics of tomato, tomatoes processing and production of puree and ketchup. Then there are described analytical methods which were used for analysing the selected parameters in ketchups. The experimental part deals with the determination of pH, total solids, organic acids, saccharides, macroelements and lycopene in ketchups. Citrate and acetate were analysed by IC. The amount of acetate in ketchups was about 5 times greater than the amount of citrate. Glucose, fructose and sucrose were analysed by HPLC. Glucose and fructose were determined in all samples but sucrose not. The ICP-OES method was used to determine four elements. Magnesium was the least represented element and sodium was the most represented element in the analysis. The amount of sodium was recalculated on the amount of salt. Lycopene was analysed by UV-VIS spectrometry. Its amount in ketchups is influenced by the variety of used tomatoes and plenty of other factors.
Miniaturized techniques for analysis of industrial yeasts

Carotenoids are natural pigments that have antioxidation and antimutagenic abilities. They are produced with the help of new technological methods. For example, carotenoid yeast Rhodotorula glutinis produces -carotene with the yield of up to 6 – 10 mg/g of the dry substance. The method of the mass spectrometry with the nanoelectrospray in the positive mode was optimized for the determination of -carotene, lycopene and astaxanthin in this project. Ionizing voltage of 4 kV and the sample flow rate of 15 – 80 nl/min through the spray silica fused capillary with the internal diameter of 25 µm were found to be the optimum parameters of the analysis. A mixture of chloroform with the addition of ammonia was used as a spray solvent for both standard and cellular samples. During the process of ionization by nanoelectrospray, -carotene and lycopene form cation radical [M] • + with the molecular mass to charge ratio (m/z) of 536, while asthaxanthin forms the protonated molecule [M + H]+ with the m/z of 597. The partial lysis of individual Rhodotorula glutinis cells was demonstrated under microscope in the organic solvents tetrahydrofuran and dimethylsulfoxide. Chloroform, acetone, acetonitrille, methanol and isopropanol did not affect the cells after a 15 min treatment.
The influence of tomato ketchupes production technology on retention of nutritionally significance ingredients

The work was focused on the verification of influence of quality of raw material and time-temperature conditions on the retention of nutritionally and sensory important components of the ketchups and other tomato based products. The goal of the theoretical part of this diploma thesis was the optimization of procedure of Czech tomato cultivar selection according to the highest refraction and intensive red colour. The influence of tomato ketchups production technology on retention of nutritionally significant ingredients was observed within the practical part. The samples of ketchups were sampled during different steps of production within one batch in company OTMA Sloko Mařatice. It was ketchup which was preserved by pasteurization in the glass bottle and ketchup which was preserved by hot filing to the plastic bottles. The content of lycopene, ascorbic acid, colour, viscosity, refraction and pH were measured in those samples. Furosine had been chosen as marker of thermal stress on material. The technologies can be compared on the basic of measured values from the view of retention or changes of nutritionally and sensory important components.
Application of spectral and non-spectral analytical methods for monitoring the quality of ketchups

The aim of the master´s thesis was to verify the applicability of elemental analysis and molecular spectrometry for examining the authenticity of ketchup. For verify of share refractometric dry matter introduced into ketchup from tomatoes was used analysis of potassium and lycopene content. Ketchups with different tomato contents were prepared and subsequently models were created, which were used to verify the proportion of refractometric dry matter introduced into tomato ketchup in ketchups from the store. The results of the work showed that the amount of lycopene is a problematic marker for verifying the authenticity of ketchup, because during the production of ketchup, lycopene is degraded.In contrast, the amount of potassium in ketchup has been shown to be a good marker for monitoring the authenticity of ketchup. The model based on the potassium content of ketchup worked reliably, the deviations obtained from the quantity of tomatoes used for the production of ketchup indicated on the packaging were less than 10%. Higher deviations were achieved in ketchups with a minimum permitted tomato content of up to 20%. The concentration of potassium in ketchup with a lower content of used tomatoes can be significantly affected by the addition of other ingredients to the ketchup (onion, garlic, carrot, apple, etc.). In addition to the ketchup production technology used, the ripeness of tomatoes also has a significant effect on the model for verifying the authenticity of ketchup.
Estery karotenoidů v potravinách a potravinových doplňcích

Carotenoids are lipophilic dyes; their production is very important for photosynthetic processes as well as for tissue protection in leaves and as attractants in fruits. They are also crucial in human/animal nutrition for their antioxidant effects. Carotenoids can be found in plant and animal tissues both in free and esterified forms. The main goal of this bachelor thesis was to establish the method of saponification of carotenoid extracts from fruits, food supplements and subsequent analysis of the presence of carotenoid esters by HPLC. Saponification was tested with sodium and potassium hydroxides. Content of carotenoid esters was detected in food samples and in selected food supplements with presumed high carotenoid content. Findings are discussed in terms of carotenoid uptake in human digestive tract.
